Reconstitution Protocol
Lyophilized peptides must be reconstituted before use in in-vitro research. Follow this protocol carefully to maintain compound integrity.
What You Need
- Bacteriostatic water (BW) — preferred for multi-use vials
- Sterile water for injection (SWFI) — for single-use reconstitution
- 1mL or 3mL sterile syringe with 27–29 gauge needle
- Alcohol swabs for stopper sterilization
Step-by-Step Protocol
- Prepare workspaceClean your work surface. Swab the vial stopper with an alcohol wipe and allow to dry for 30 seconds.
- Draw bacteriostatic waterUsing a sterile syringe, draw the desired volume of BW (use the calculator above to determine amount).
- Inject slowlyInsert the needle through the vial stopper at an angle. Inject the water slowly down the inner side of the glass — do not spray directly onto the lyophilized cake.
- Gentle swirl onlyGently swirl the vial in a slow circular motion. Do not shake vigorously — this can denature peptide structures.
- Inspect and labelThe solution should be clear. If cloudy or particulate, do not use. Label the vial with compound name, concentration, date, and initials.
- Refrigerate immediatelyStore reconstituted solution at 2–8°C (standard refrigerator). Use within 30–60 days for optimal research integrity.

What is Lyophilization?
Lyophilization (freeze-drying) is the gold standard for preserving analytical-grade peptides. The process removes water through sublimation under vacuum, converting the aqueous peptide solution directly from a frozen state to a dry powder — bypassing the liquid phase entirely.
